δ Chain of TCR Sequencing Service

As part of the T cell receptor (TCR) complex that makes up the γδ T cells, sequencing of the δ chain is important for revealing the mechanisms of many diseases, such as infectious diseases, organ transplantation, cancer, and autoimmune diseases. Special TRD Locus

The genes for TCR are located at four discrete loci on the chromosomes, TRA, TRB, TRG, and TRD. The TRA and TRD genes are located at a single chromosomal location, and the TRD locus is nested within the TRA locus. The genomic organization of the TRA/TRD locus is known for both humans and mice.

The human TRA/TRD locus spans 1.1 Mb, and five of the V genes have been found to rearrange in the TRA and TRD chains (TRAV/DV genes). Four TRDV genes, one of which is located between the TRAV genes, three TRDD genes, four TRDJ genes, and one TRDC gene, plus a TRDV gene with reversed transcriptional orientation, form a TRD motif nested within the TRA motif.

The mouse TRA/TRD locus spans 1.7 Mb and is predominantly occupied by 132 TRAV genes, including 9-10 TRAV/DV genes. The TRD genes include five TRDV, two TRDD, two TRDJ, and one TRDC gene, followed by a TRDV gene with inverted transcriptional orientation.

Rationale of the δ Chain Sequencing and Analysis

Insight into the formation of the γδ TCR complex and the mechanisms of ligand and factor activation facilitates our understanding of disease and immune processes. Notably, the V gene in γδ T cells is not only related to their function and individual development, but also to their tissue localization. In the healthy liver, spleen, lymph nodes, and lung, the complexes of γδ TCR are oligoclonal distribution.

For example, in healthy individuals, lung γδ T cells are enriched for Vδ2+ T cells, whereas patients with active tuberculosis have been reported to have increased numbers of Vδ1+ T cells showing skewed TCR rearrangement.

The TCR repertoire of γδ T cells is very different from that of αβ T cells. Although theoretically the TCR complex diversity of γδ T cells is estimated to far exceed that of αβ T cells, γδ T cells are still understood to be more invariant T cells that use only a limited set of γδ TCRs. High-throughput TCR repertoire sequencing has contributed to the understanding of γδ T-cell repertoire diversity and the dynamics of the human γδ TCR repertoire in health and disease.
