Introduction to the Mucosal Immune System
The mucosal surface is continuously exposed to the external environment and therefore represents the largest lymphoid organ in the body. Within the mucosal immune system, gut-associated lymphoid tissues (GALTs), including Peyer's patches and isolated lymphoid follicles, play an important role in the induction of antigen-specific immune responses in the gut. GALTs have unique organogenesis characteristics and interact with dendritic cells and T-cell networks to both induce and regulate IgA responses and oral tolerance.

Innate lymphocytes (ILCs) are abundant in mucosal tissues and are involved in tissue homeostasis and barrier function. To exemplify the determination of their differentiation pathways, ILCs in the human fetal gut are analyzed using mass spectrometry and 34 distinct populations are distinguished. The results identify a smooth phenotypic transition from cells within the Lin-CD7+CD127-CD45RO+CD56+CD8a- cluster to CD45RA+ NK cells and CD127+RORγt+ ILC3-like cells, revealing potential differentiation trajectories.
